乙醇對心肌梗死大鼠冠脈血管新生的影響

復里學報(醫(yī)學版Fudan Univ J Med Sci 2014 Jul, 41(4)乙醇對心肌梗死大鼠冠脈血管新生的影響屈福超王愛紅孫永樂苑海濤張玉英4(山東大學附屬省立醫(yī)院心內(nèi)科濟南250012)【摘要】目的探討乙醇對大鼠急性心肌梗死后冠脈側(cè)支循環(huán)形成的影響及其機制。方法取雄性SD大鼠隨機分成4組:假手術組空白對照組,小劑量乙醇組(50%乙醇,0.5g·kg1·d1)及大劑量乙醇組(50%乙醇5g·kg·d-1)。制作心肌梗死模型每組各8只,于術后第3天處死,留取梗死邊緣區(qū)存活心肌,采用免疫組織化學SP法檢測梗死邊緣區(qū)心肌內(nèi)微血管密度( microvascular density,MvD),ELA法測定血管內(nèi)皮生長因子( vascular endothelial growth factor,vEGF及內(nèi)皮抑素水平。通過方差分析研究各組間MVD及VFGF內(nèi)皮抑素表達水平有無差異。結(jié)果與假手術組相比,空白對照組MVD明顯增加(P<0.05),VFGF、內(nèi)皮抑素水平明顯升高(P<0.05);小劑量乙醇組MVD較空白對照組明顯增加(P<0.01),vEGF明顯升高(P<0.05),內(nèi)皮抑素濃度明顯降低(P<0.05);而大劑量乙醇組MVD則明顯減少(P<0.01),VEGF水平與空白對照組相比差異無統(tǒng)計學意義,而內(nèi)皮抑素明顯升高(P<0.05)。結(jié)論乙醇能夠改變心肌梗死后缺血心肌內(nèi)VEGF和內(nèi)皮抑素的表達,少量乙醇攝人可以通過增加EGF的表達促進缺血心肌內(nèi)的血管新生過程,而大量乙醇攝入則提高內(nèi)皮抑素的表達而抑制缺血心肌的血管新生過程【關鍵詞】乙醇;心肌梗死;微血管密度(MVD);血管內(nèi)皮生長因子(VEGF);內(nèi)皮抑素;大鼠【中圖分類號】R542.22【文獻標志碼】Adol:10.3969/ I. issn.16728467.2014.04.018Effect of ethanol on the coronary angiogenesis afteracute myocardial infarction in ratsQU Fu-chao, WANG Ai-hong, SUN Yong-le, YUAN Hai-tao, ZHANG Yu-yingaDe partment of Cardiology, Shandong Provincial Hospital, Shandong UniversityJinan 250021, Shandong Province, China)(Abstract] Objective To investigate the effect of ethanol on coronary angiogenesis after acutemyocardial infarction in rats.
